I just got a 1 TB HDD to hook up to my DVR (HR20-700). Does this disk need to be initialized and formatted before use, or do I just hook it up just as it comes from the manufacturer? It is a Seagate Barracuda 7200.11

Nowhere do you say what enclosure you are using; you DO know that putting it into the actual DVR DOES violate a couple of the agreements you have with DirecTV, unless you OWN the unit ((~$800), and I would doubt that (you are paying the $4.99/month LEASE fee, right?).

Connecting up to the eSATA port, it all depends on the external boxes interface, NOT the interface on the drive itself. The drive connects to the internal port on the external interface, and the external port on the box (eSATA) connects to the eSATA port on the HR20-700.

Simple.
